Abstract

1.
1. This report describes a simple and efficient procedure for the isolation of plasma membrane from guinea pig peritoneal macrophages. The use of polycationic beads (Affi-gel 731 beads) facilities rapid and high-clear separation of plasma membrane within 30 min.
2.
2. The final plasma membrane coated beads fraction has high specific activities of marker enzymes with little contamination with mitochondrial, lysosomal or cytoplasmal markers.

Interleukin-17A: Potential mediator and therapeutic target in hypertension

γδ lymphocytes are involved in the immune response against fungi and pathogens, as well as in autoimmune diseases.70,71 These are unconventional T cells, which recognize many microorganisms and transform host cells, acting as the first line of defense in peripheral tissues.72,73 The IL-17A produced by these cells is mainly involved in antifungal immunity, such as the response to infection by Candida albicans70; and in the initial stages of autoimmune pathologies.74
Interleukin-17A (IL-17A) is a proinflammatory cytokine produced by cells of the immune system, predominantly Th17 and γδ lymphocytes. In this paper, we review the role of IL-17A in the pathogenesis of hypertension and in target organ damage. Preclinical studies in mice have shown that systemic adminstration of IL-17A increases blood pressure, probably by acting on multiple levels. Furthermore, IL-17A plasma concentrations are already elevated in patients with mild or moderate hypertension. Many studies in hypertensive mice models have detected IL-17A-producing cells in target organs such as the heart, vessels and kidneys. Patients with hypertensive nephrosclerosis show kidney infiltration by Th17 lymphocytes and γδ lymphocytes that express IL-17A. In addition, in experimental models of hypertension, the blockade of IL-17A by genetic strategies or using neutralizing antibodies, disminished blood pressure, probablyby acting on the small mesenteric arteries as well as in the regulation of tubule sodium transport. Moreover, IL-17A inhibition reduces end-organs damage. As a whole, the data presented in this review suggest that IL-17A participates in the regulation of blood pressure and in the genesis and maintenance of arterial hypertension, and may constitute a therapeutic target of hypertension-related pathologies in the future.
La interleuquina 17A (IL-17A) es una citoquina proinflamatoria producida por células del sistema inmune, sobre todo por los linfocitos Th17 y los linfocitos γδ. En este trabajo, revisamos el papel de IL-17A en la patogenia de la hipertensión y de la lesión en órganos diana. Estudios en ratones han demostrado que la IL-17A aumenta la presión arterial, probablemente por acciones a varios niveles. Además, las concentraciones plasmáticas de IL-17A están ya aumentadas en pacientes con hipertensión arterial ligera o moderada. Estudios preclínicos sobre hipertensión arterial han detectado células productoras de IL-17A en órganos diana, como corazón, vasos y riñón. En pacientes con nefroesclerosis hipertensiva existe infiltración del riñón por linfocitos Th17 y linfocitos γδ que expresan IL-17A. Además, en modelos experimentales de hipertensión el bloqueo de IL-17A, mediante estrategias génicas, o utilizando anticuerpos neutralizantes, disminuye la presión arterial por acciones sobre la pared vascular y el transporte tubular de sodio y disminuye la lesión en órganos diana. En conjunto, los datos presentados en esta revisión sugieren que la IL-17A participa en la regulación de la presión arterial y en la génesis y mantenimiento de la hipertensión arterial, pudiendo constituir una diana terapéutica en el futuro.
Identification of a putative cell receptor for human cytomegalovirus

Proteins from a variety of cell types were separated using SDS-PAGE, transferred to nitrocellulose filters, and probed with intact human cytomegalovirus (HCMV). Virus bound to cell proteins was detected directly using ¹²⁵labeled HCMV or indirectly using an immunologic assay with a secondary antibody labeled with ¹²⁵I. Using this approach proteins of molecular weights 32 and 34 kDa were identified that would bind HCMV and were present on T4+ and T8+ lymphocytes, a B lymphoblastoid cell line, and human diploid fibroblasts. Binding of labeled virus to cells could be blocked by the addition of unlabeled homologous virus. Treatment of virus with NP40 to remove the virus envelope blocked binding to cellular proteins. Both the 32- and 34-kDa proteins could be copurified with cellular membranes. HCMV bound equally well to T4- and T8+ cells as well as to either replicating (PHA or interleukin-2 stimulated) or resting lymphocytes. Interestingly, there appeared to be a single binding site for HCMV on human fibroblasts (34 kDa). The results support the idea that there is a receptor for HCMV present on the surface of human lymphocytes and fibroblasts.
Phospholipase A<inf>2</inf> in macrophage plasma membrane releases arachidonic acid from phosphatidylinositol

A high level of arachidonic acid release from [2-¹⁴C]arachidonylphosphatidylinositol (PI) was observed at neutral pH (6.0–7.0) in the presence of purified plasma membranes of guinea pig peritoneal macrophages. This activity was at least 10-fold higher than that with arachidonylphosphatidylcholine (PC) or phosphatidylenatholamine (PE) as substrate. The accumulation of [¹⁴C]diacylglycerol and [¹⁴C]phosphatidic acid was not detected at any time, and arachidonic acid release from [¹⁴C]arachidonyldiacylglycerol was not detectable either. The data suggest that arachidonic acid release from PI may not occur via the phospholipase C pathway. In this paper, we demonstrate the possibility that arachidonic acid release from PI at neutral pH in the macrophage plasma membrane is dependent on the action of phospholipase A₂ (EC 3.1.1.4) -like activity. The maximum arachidonic acid release was dependent upon both pH and substrate. Particularly, the activity of arachidonic acid release from PI at neutral pH was very high compared with that from PC or PE. We suggest that phosphatidylinositol phospholipase A₂ (EC 3.1.1.52) may play an important role in providing arachidonic acid for subsequent metabolic activity in the macrophages.
Recognition of acidic phospholipase A2 activity in plasma membranes of resident peritoneal macrophages

Phospholipase (PLase) activities in the plasma membrane of guinea pig peritoneal macrophages were studied, as these enzymes having such activity may be candidates for the release of arachidonic acid (AA) from phosphatidylcholine (PC). An AA release system operating at acidic pH was identified in the macrophage plasma membrane and characterized. This membrane-bound acidic PLase A₂ had an optimum pH at 4.5, and enzyme activation was observed in Ca⁺⁺-free medium; but the maximum activity was found at 0.5 mM Ca⁺⁺ concentration. The Km value for Pc of acidic PLase A₂ was 4.2 μM, and a Michaelis-Menten relationship was evident. Calcium might act as a cofactor at some intermediate step during the activation of acidic PLase A₂ in light of the uncompetitive manner of Ca⁺⁺ action. Furthermore, the release of [³H]-AA from preradiolabelled macrophage plasma membranes occurred with the addition of Ca⁺⁺ at pH 4.5. These data suggest that the acid PLase A₂ is a component of the plasma membrane and is not due to lysosomal contamination since membrane-bound acidic PLase A₂ properties are opposite to those found for lysosomal PLase A₂.
